Craniofacial development: Knockout mice take one on the chin
نویسندگان
چکیده
Face and head morphogenesis begin early in development, when the brain is beginning to form, and are highly dependent on patterning information provided by emigrant cranial neural crest cells [1]. These cells generate most of the bones and cartilage in the head, and instruct paraxial mesoderm in the formation of appropriate musculature [2]. Molecular cues that differ spatiotemporally along the rostrocaudal axis of the early brain plate affect emerging cranial neural crest cells and predetermine some aspects of their fate, such as whether they generate first or second branchial arch skeletal elements.

G-Protein α-Subunit Gsα Is Required for Craniofacial Morphogenesis.
The heterotrimeric G protein subunit Gsα couples receptors to activate adenylyl cyclase and is required for the intracellular cAMP response and protein kinase A (PKA) activation. Gsα is ubiquitously expressed in many cell types; however, the role of Gsα in neural crest cells (NCCs) remains unclear. Here we report that NCCs-specific Gsα knockout mice die within hours after birth and exhibit dram...

Holoprosencephaly is a common developmental disorder in humans characterised by incomplete brain hemisphere separation and midface anomalies. The etiology of holoprosencephaly is heterogeneous with environmental and genetic causes, but for a majority of holoprosencephaly cases the genes associated with the pathogenesis could not be identified so far. Here we report the generation of knockout mi...

H2-EB1 Molecule Alleviates Allergic Rhinitis Symptoms of H2-Eb1 Knockout Mice
Background: H2-EB1 molecule which is the homolog of Human HLA-DRB1 is proposed to be associated with allergic rhinitis (AR). Construction of H2-Eb1 knockout animal models provides a tool to elucidate the role of H2-EB1 and AR pathogenesis. Objective: To establish the H2-Eb1 knockout model and investigate the H2-EB1 functions in H2-Eb1 knockout mice as a model of AR. Methods: The Cre/Lox...
متن کاملInfluence of growth hormone on the craniofacial complex of transgenic mice.
Growth hormone (GH) secretion affects bone and cartilage physiology. This study investigated the effect of GH on the size of the craniofacial structures and their angular relationship. Three different models of mice with a genetically altered GH axis were used: GH excess (giant), dwarf GH antagonist (dwarf-Ant), and dwarf GH receptor knockout (dwarf-KO) mice. Each model was compared with the co...
